General Information
Introduction to Toxins
Aconitum alkaloids are highly toxic diester diterpenoid alkaloids contained in Aconitum plants (such as Aconitum
chuanensis, Aconitum sinensis, and Aconite, etc.), including aconitine, subaconitine and neoaconitine. This type of
component is not only the main active substance of traditional Chinese medicine but also the source of poisoning.
The therapeutic dose is close to the toxic dose, and 0.2 milligrams of aconitine can cause poisoning.
Application
PriboStrip TM aconitines qualitative rapid test strip is used for the rapid detection of aconitine residues in samples
related to wine-making. The entire detection process only takes 10 minutes and is suitable for on-site rapid
screening by various enterprises and testing institutions.
Testing Principle
The reaction zone of the test strip is coated with two lines: the test line (T) and the control line (C). The C line
serves as the quality control line, while the T line serves as the detection line and is coated with a ACO–BSA
conjugate. The colloidal gold microwells contain a conjugate of anti-ACO monoclonal antibodies labeled with
colloidal gold (Ab-Gd).
If the target analyte (ACO) is not present in the sample, the Ab–Gd conjugates specifically bind to the ACO–BSA
conjugate at the T line, resulting in the appearance of a red line. The remaining Ab–Gd conjugates continue to
migrate to the C line, where they also form a red line.
If ACO is present in the sample, the free ACO competes with the ACO–BSA conjugate for binding to the Ab–Gd
conjugates. As a result, the color intensity of the T line decreases or disappears, while the C line remains visible.

Note: Recovery to room temperature and mix well before use. If solid precipitates appeared, please redissolve it at
25~30 ℃.
Sample extract solution was prepared
50% ethanol water extract: take 500 mL of pure water, add 500 mL absolute ethanol (AR grade), mix and seal at
room temperature.
Notes
1. The optimum operation temperature is 20-25℃.
2. The strip is disposable and cannot be reused.
3. Do not use expired test strip.
4. Do not touch the white membrane in the center of strip.
5. Do not reuse the straw provided in case the cross-contamination.
6. Do not mix the strips and microporous reagent with different lot numbers.
7. If test the standards, please request for the corresponding buffer. Tap water, distilled water, deionized
water should not be used as the negative control.
8. The solid impurity particles in the sample will lead to false positive results. Please discard the visible
particles in the samples. (If necessary, suggest you centrifuge the initial sample and collect the supernatant
as test sample).
9. Aconitines is a highly toxic substance, please operate with gloves.
10. Please contact the supplier if any problem during testing.
11. The test results are for reference only. For confirmation, please refer to the relevant national standard
methods.
Sample Preparation
1. Soaked alcohol sample (detection limit:15 PPB)
Take 0.1 mL of the soaked wine and mix it evenly with 0.5 mL of the sample dilution solution. This is the test
solution.
2. Extraction raw material sample (detection limit: 60ppb)
Take 1 g of the homogenized test sample, add 3 mL of 50% ethanol, and transfer it to a 10 mL centrifuge
tube. Shake and mix well for 5 minutes. Let it stand or centrifuge for 1 minute. Take 0.1 mL of the
supernatant and mix it evenly with 0.5 mL of the sample diluent. This is the test solution.
3. Serum (detection limit: 6ppb)
Take 0.1 mL of serum and mix it evenly with 0.5 mL of the sample dilution solution. This is the test solution.
Test Procedures
When removing the test card, do not touch the reaction area or the sample area to ensure the accuracy of
the test.
1. Please read the manual thoroughly before testing. Recovery the test strip and test solution to room
temperature (20-25°C) before use.
2. Take out the strips from the original packaging bag.
3. Take 100 μL of the solution to be tested and add it to the sample well of the test card. Incubate for 10
minutes.
4. Read the results at 15 minutes; otherwise, the results may be invalid.

Negative(-): T line is darker than C line. This result means that the concentration of aconitine was
lower than the detection limit or there was no aconitine residue.
Positive(+): T line is lighter than C line/the same as C line/T line has no color. This result means that the
concentration of aconitine was higher than the detection limit. The lighter of T line means the higher
content of aconitine residue.
Invalid: The C line shows no color. The operation was performed improperly, or the test strip has failed.
Please repeat the test with a new strip.
Validity and Storage Conditions
Avoid direct light and store at 2 - 8 ℃, do not freeze.
Lot number and Expiry date
The lot number and expiration date are printed on the outside of the package.
